What Is Their Main Difference?

Both are exclamations. However, 'yay' indicates excitement for what is going to happen, whereas, 'nay' is a negative exclamation that shows disagreement.

Differences

'Yay' is a positive exclamation that shows excitement for what is about to happen, while, 'nay' shows disagreement and is negative. Look at the examples below:

Yay! We're going to London.

They all yelled: 'Nay'.

Similarities

Are They Interchangeable?

Since they are antonyms, they cannot be used interchangeably. Compare:
