What Is Their Main Difference?

The main difference between 'although' and '*in spite of' is in their parts of speech. 'Although' is a conjunction while 'in spite of' is a preposition.

Parts of Speech

As mentioned earlier, 'although' and 'in spite of' are different in their parts of speech.
'Although' is a conjunction. It is classified as a subordinating conjunction. 'In spite of,' however, is a preposition. Take a look at the folloing examples:

Although he hated the song, he played it everyday.

In spite of the fact that he hated the song, he played it everyday.

Position in a Sentence

'Although' and 'in spite of' can be used at the beginning and the middle of a sentence. Watch:

Although he had a lack of experience, he got the job.

He got the job although he had a lack of experience.

He got the job in spite of his lack of experience.

In spite of his lack of experience, he got the job.

Frequency

'Although' and 'in spite of' are more commonly used in written English than spoken English.

The example should read: Despite his lack of experience, he got the job. This example goes from bad to good. This contrasts with in spite of, which goes from good to bad: In spite of her being a genius, she played a lousy game of chess.
